This kit utilizes isothermal nucleic acid rapid amplification technology. At a constant temperature (typically 39–42°C), auxiliary proteins and single-strand binding proteins facilitate the formation of a complex between recombinase and primers. The complex then searches for and binds to the target homology domain. This binding forms a D-loop region at the homologous site, initiating strand exchange. As the recombinase dissociates, polymerase binds to the 3' end of the primer, beginning chain extension. By utilizing the nfo enzyme activity, a specific molecular probe is introduced based on the template, and colloidal gold technology (sandwich method) is employed to detect the final result.

【Primer design】
It is recommended to use primers with a length of 30–35 nt. Shorter primers may reduce amplification speed and detection sensitivity. The downstream primer should have a modification group (commonly biotin) at the 5' end.Primer design should avoid secondary structures that may impede amplification. The recommended amplicon length is 150–500 nt.
【Colloidal gold probe design】
The probe sequence, complementary to the target fragment, should be 46–52 nt in length, designed between the upstream and downstream primers.
- An antigen marker (typically FAM) is attached to the 5' end.
- A dSpacer (tetrahydrofuran, THF) is labeled in the middle of the probe, between the 5' and 3' ends, as the recognition site for nfo.
- A modification group, such as an amine, phosphate, or C3-spacer, is labeled at the 3' end.

- The kit is available in two types - DNA and RNA. The RNA type includes reverse transcriptase, allowing amplification with RNA, dsDNA, and ssDNA as templates. The DNA type can only amplify with dsDNA as the template.
